Isolation and Purification of Pregnacy-Specific-Protein (SP©û)


Abstract
Pregnancy-specific-1-glycoprotein (SP,) is one of the pregnancy specific protein, isolated by Bohn at 1972 from placental tissue. It¢¥s amount wtihin pregnant women¢¥s sera have a strong positive association with fetal body weight and negative association with degree of severe preeclampsia, which could induce retardation of fetal growth.
It¢¥s sedimentation coefficient is 4.6 S, composed of two subunits of about 2.8 S. Molecular weight 90,000. Isoelectric points 3.9. It is composed of hexose 10.8%, hexosamine 9.9¡Æ¢¥0, fucose 67o, sialic acid 6. 8% and its total carbohydrate composed of 28.17o of molecular weight, i. e. glycoprotein.
SP, was known as synthesized in trophoblasts (Bohn) and partly in fetal tissue(Park and Kim)
and it¢¥s half life is 30 to 40 hours. Its physiologic action in vivo is not well known, yet, but it was suggested that it might have strong positive association with placental condition, i. e. placentalfetal stability.
In order to purification of SP,, the authors collected and pooled sera from 230 normal pregnant women from September 1980 to March 1981. After DEAE-cellulose colum chromatography, balan ced with sodium acetate buffer, pH 4.7, the sample were electrophoresed on the starch block, with barbital buffer, pH 8.6, for 22 hours with 6 v/cur. After electrophoresis, salting out with 2.6 M ammonium sulfate solution to isolated transferrin were performed,
The supernatant was confirmed as SP, by immunodiffusion and immunoelectrophoresis.
